How to read and verify a peptide COA

A peptide Certificate of Analysis is only as useful as what it actually reports. A complete COA identifies the exact lot or batch it describes, names the independent, third-party laboratory that ran the tests, and shows the underlying data — not just a headline number. The two analyses that matter most are the HPLC chromatogram, which measures chromatographic purity, and the mass spectrum (ESI-MS or LC-MS), which confirms the compound's identity by molecular weight. Reading a peptide COA well means checking that both are present, with peak integration and area% behind any purity figure.

HPLC and mass spec answer different questions, and a peptide purity certificate should include both. HPLC tells you how pure the sample is relative to other species that absorb light; mass spec tells you whether the main peak is actually the peptide you ordered. A number like "99% pure" with no chromatogram, no integration, and no spectrum is a claim, not evidence. Third-party peptide testing — where an accredited lab independent of the seller runs the assays — is what lets you verify a peptide COA rather than take it on faith. Supporting tests such as water content (Karl Fischer), endotoxin (LAL), and counter-ion / net-peptide content round out a rigorous report.
